We show that the essential protein translation machinery enzyme called lysyl tRNA synthetase (KRS) from different helminths can be inhibited by the same drug cladosporin. This active site binding inhibitor disables with equal potency both the African eye worm Loa loa's KRS and the snail fever causative agent S. mansoni's KRS. The structural and biochemical data suggest that derivatives of cladosporin may be promising new chemical series that can be rationally developed as new drugs against several neglected tropical diseases that are caused by eukaryotic parasites.
