Reviewer #2: The authors conducted a statistical analysis using Distributional Synthetic Controls (DSC) approach to evaluate the spatiotemporal heterogeneity in the intervention effects of the Incompatible Insect Technique coupled with the Sterile Insect Technique (IIT-SIT) in Singapore. This looks like a follow-up study based on their own prior study of Ref [4].

Their previous study Ref [4] considered average mosquito abundance over time. This new study did some extension on the temporal variation on intervention effects. Using data from 57,000 traps, the authors provided a more granular evaluation of intervention efficacy.

(1) I have some concern on the novelty of this new work. It looks like most key findings have been published in their previous work Ref [4]. I am not sure if the current study gives more insights on the IIT-SIT interventions.

(2) The presentation is difficult to follow. The authors are good at summarizing technical details. However, this makes readers feel distraction as it’s not easy to get their points. To be honest, the format of this paper is more like a technical summary instead of research article. I would suggest the authors carefully edit the manuscript according to the format and guidelines of PLoS Computational Biology.

(3) It is unclear to me why the DSC approach is appropriate and the best approach to evaluate the causal relationship in the study setting. Given the presence of other confounding factors such as climate factors, intervention measures induced by COVID control policies, human population awareness of infectious pathogens, I’m nervous about the claim on the inference of causal relationship.

(4) Line 158, the authors mentioned “We used the procedure of Li and Tolhurst by replacing uniform random sampling of quantile functions to beta distribution random sampling [16].” Given that the choice of sampling distribution can impact the estimated synthetic counterfactual, the authors are recommended to perform a sensitivity analysis to see if the findings remain robust if a standard uniform distribution or different Beta parameters are used.

(5) Line 150, "Only the top 40 control sectors which were most correlated to a specific intervention sector were used for fitting the DSC”. It is unclear whether this "top 40" threshold might introduce a selection bias that favors control sectors with similar local environmental shocks rather than those representing the broader counterfactual trend.

Singapore. All data access enquiries should be directed to the agency". There is no any plan or GitHub page addressing the sharing of data and code. It looks impossible to replicate the study.

Reviewer #3: My very minor comment is I think the abstract and author summary can ‘sell’ the developed method a bit better rather than only focus on how effective the treatment was – how this newly developed method differs to the previous one and what’s been added to the tools of intervention effectiveness evaluation available so far.

This part in SI is incomplete:

However, we found similar trajectories in treatment effect. During the first 52-week treatment, average IE in core areas increased from 28.09% (95%CI: 17.78% – 40.81%) to 62.63% (95%CI: 48.41% – 81.78%), while average IE in buffer areas increased from 20.71% (95%CI: 11.52% – 29.65%).

For example Figure 2 and 4 have confidence intervals (I assume) around each line as a shaded region, but the caption and figure does not state what the shaded region is, or what confidence level it represents (e.g. 95% confidence intervals?). Figure 5 has box and whiskers, but their meaning has not been explained in the caption.
  • Figure 2B has the shaded region missing on some of the lines? Figure 2D has a different colour shaded region around the bottom most line compared to the other panels (and the legend).
