Phosphorylation-dephosphorylation cycles can generate non-monotonic input-output relationships.

Here the allostery of the substrate inhibits further modifying reactions by disfavouring binding of the kinase when the substrate is phosphorylated and disfavouring binding of the phosphatase when the substrate is dephosphorylated.
