Single source specimens for plants (A), fungi (B), bacteria (C), and arthropods (D) underwent melt curve analysis for specificity testing of each primer pair. Each trace is a different sample. The y-axis is the derivative reporter fluorescence (-Rn’) and the x-axis is temperature (°C). The peak of the melt curve is the temperature at which 50% of DNA is double stranded and 50% is single stranded.
