Immunofluorescent analysis of podoplanin-regulated ezrin and Rho-GDI presence in plasma membrane of MSU1.1 and HS578T cells.

Podoplanin-negative and podoplanin-overexpressing MSU1.1 and HS578T cells were immunofluorescently labelled to visualize distribution of ezrin and Rho-GDI proteins. Based on plasma membrane and cytoplasm co-staining with WGA-Alexa Fluor 647 and CTV, respectively, ezrin (A) and Rho-GDI (B) content in plasma membrane was quantified with confocal microscopy and image analysis using ImageJ software. Data are presented as mean fluorescence intensities in identified plasma membrane regions (single dots). Data were analyzed using the Mann–Whitney test, *p<0.05, **p<0.01, ***p<0.001, ****p<0.0001.
