Dual inhibition of Fes and Flt3 tyrosine kinases potently inhibits Flt3-ITD+ AML cell growth

A) The Flt3-ITD+ cell lines MV4-11, MOLM-13, and MOLM-14, along with THP-1 (wild-type FLT3) were incubated with each of the kinase inhibitors shown over a range of concentrations, and cell viability was measured 72 h later using the CellTiter-Blue Assay (see Materials and methods). Raw fluorescence data were corrected for background and then normalized to cultures grown in the presence of the DMSO carrier solvent. Each assay condition was performed in triplicate, and results are presented as the mean of the normalized values ± the SD. The resulting dose-response curves were fitted by non-linear regression analysis, and the resulting IC50 values are shown in Table 1.
