The solid smooth lines represent the best fit of the data for the IC50 values as 7.0 ± 2.9 mM (in the presence of 50 μM NAD+) and 8.9 ± 3.3 mM (in the presence of 5 mM NAD+), respectively. (B) Dose-response of SIRT5 desuccinylation activity in the presence of varying concentrations of isonicotinamide (from 0 to 15 mM) and NAD+ (50 μM and 5 mM). The solid smooth lines represent the best fit of the data for the IC50 values as 11.1 ± 2.2 mM (in the presence of 50 μM NAD+) and 17.1 ± 4.0 mM (in the presence of 5 mM NAD+), respectively.
