Lipophilic Prodrugs of FR900098 Are Antimicrobial against Francisella novicida In Vivo and In Vitro and Show GlpT Independent Efficacy

The antimicrobial effect of fosmidomycin, FR900098, compound 1 and compound 2 against F. novicida.
The EC50s (the concentration at which 50% of bacterial growth is inhibited) of fosmidomycin (A), FR900098 (B), compound 1 (C), and compound 2 (D) were determined as previously described. Briefly, serial dilutions of the antibiotic were performed in a 96-well plate format to a defined concentration of bacteria. The percent growth compared to wells with no antibiotic was graphed using GraphPad Prism 4.0. The EC50 of fosmidomycin was 3.6±0.2 µM. The EC50 of FR900098 was 23.2±1.2 µM. The EC50 of compound 1 was 45.2±3.7 µM and the EC50 of compound 2 was 481±44 µM.
