Dr. Mohammed Rohi Khalil et al. presents data suggesting that the intrapartum PCR assay performs better than the antepartum culture for identification of GBS vaginal carriers during labor.
Nevertheless, the authors compare the PCR assay with culture in Granada Medium but they omit two key issues (1,2). First, they incubate Granada medium plates aerobically in an atmosphere containing CO2, overlooking the fact that Granada plates must be incubated in anaerobiosis. And two, surprisingly, they plated the vaginal and rectal swabs in the Granada plates omitting the mandatory step of enrichment in selective broth.
Hence, the omission of these key requisites for correct detection of GBS by culture indicates that the conclusions of this paper should be taken with caution.
