Lifespan extension of class 1 daf-2 mutants by DR

Surprisingly, DR had no effect on lifespan of daf-2(e1368) adults (Fig. 5A). This finding suggests that DR and daf-2(e1368) may extend adult lifespan through overlapping mechanisms that are not additive.

The level of dietary restriction that maximizes lifespan varies for different genotypes. Therefore, before the conclusion that DR does not extend the lifespan of class 1 mutants can be drawn, it would be beneficial to test the lifespan of the e1368 worms across a range of bacterial dilutions (or even dietary deprivation). It may be that although DR via eat-2 mutation does not extend the lifespan of these worms, their lifespan can be increased by DR if the food intake level is optimized.

Yes, you have raised a good point. It is difficult to know whether the conditions are optimized for DR, or for IIS reduction for that matter. However, the finding that these daf-2 alleles differed in their responses to eat-2 induced DR is important evidence for some sort of interaction between these pathways.
