I wonder why the authors did not measure LpPLA2 activity or mass in their samples and correlate the data with the methylation pattern? As pointed out in the limitation section the authors worked with a mixture of cells and to my knowledge most of them will never secrete LpPLA2. Perhaps they should repeat the work with isolated monocytes.
At this point I miss the functional connection between the methylation of this part of the promoter and CHD.

The purpose of this pilot study is to test whether the promoter methylation as a biomarker works in the prediction of the risk of CHD by using the peripheral blood. And you are right about using the monocytes to explore the functional link between methylation and CHD. And it will be even better adding the LpPLA2 actiivity in the serum. We thank you very much for your great suggestion, and will try our best to collect all-angle data in the coming projects.
