Anti-EphB2 autoantibody

Dear Authors,

We read this article with great interest because we had also reported anti-EphB2 autoantibody in an SLE patient who manifested acute necrotizing encephalopathy [1]. We have been trying to identify autoantigens expressed on the cell surface, and had established the SARF method using retroviral vector and flow cytometry [2]. The anti-EphB2 autoantibody from the patient reacted with the extracellular domain of EphB2, which was confirmed by flow cytometry. We had measured autoantibodies against extracellular domain of EphB2 in 47 SLE patients, but none of them showed binding activity to the extracellular domain of EphB2 [1]. Therefore, we speculated that anti-EphB2 antibody had potential to induce vascular lesion in the acute necrotizing encephalopathy.
In this article, authors utilized ProtoArray analysis and showed that anti-EphB2 autoantibodies were detected in 56 % of SLE patients, and most of them recognized the intracytoplasmic region of EphB2. Furthermore, they revealed that 34% of SSc patients also possessed anti-EphB2 autoantibodies. These data are pretty exciting and we would like to know whether these patients have anti-EphB2 autoantibodies against extracellular domain because such autoantibodies can directly induce vascular inflammation.

Dear colleagues,

We are glad you found our recently published results exciting [1]. We also read with a great interest your paper reporting anti-EphB2 autoantibodies (AAb) in a patient with neuropsychiatric Systemic Lupus Erythematosus (SLE) who manifested acute necrotizing encephalopathy [2]. We did not mention your work in our manuscript precisely because your work was relative to AAb recognizing the extracellular domain of EphB2. In our study, all the patients with SLE who had anti-EphB2 AAb had them recognizing the intracytoplasmic region of EphB2 and precisely the tyrosine kinase domain.
Indeed on the version 5.0 of Invitrogen Human protein microarrays (Invitrogen, Carlsbad, CA, USA), the 3 proteins spotted on the nitrocellulose-coated glass slide called “Ephrin type-B receptor 2”, represented only the tyrosine kinase domain more or less truncated (see www.thermofisher.com for protein content list 5.0). Therefore we have never tested whether our sera samples have AAb against the EphB2 extracellular domain. We agree that it would be very informative to do it.
On the other hand, we would be very curious to know whether the 47 sera from Japanese patients with SLE, previously tested in your study for their binding activity to the extracellular domain of EphB2 and found negative, would be positive for AAb against the tyrosine kinase domain of EphB2 and particularly against the peptide 7 we describe as being the major epitope in Lupus.
