How is the percentage of false positive cases on deep sequencing?

Dear Dr. Tallini and Dr. Biase:

Congratulation for your work. It is a useful tool on patient screening for target therapy. I just have a few comments about the work:
1. What is the percentage of false positive cases from your deep sequencing using NGS? Should you have a positive and negative control or use other methods to confirm your positive result by deep sequencing (5% mutation DNA)?
2. If the percentage of mutant EGFR is very low, is there any different on TKI response from the patients with higher copy number? Based on some clinical data, TKI response is not just relative to the status of EGFR mutation, but also to the copy number of the mutant gene.
