Comments on anti-P-T389-S6K1 used by the authors

The authors stated that the previously described activation of S6K1 in response to H2O2 by other authors, including our work (J Biol Chem, 2012. 287(4): p. 2632-42), in different manuscripts and journals is a misinterpretation of the non-specific detection of proteins p90 RSK and MSK by the antibody most broadly used in the literature, the anti-P-T389-S6K1 (1A5), Mouse mAb #9206 (Cell signaling). So, they encourage researchers using anti-P-T389-S6K1 antibody to confirm the correct size of the band detected.

Competing interests declared: However, anti-P-T389-S6K1 antibody (1A5), (Mouse mAb #9206 from Cell signaling) was never used in our work; instead we used anti-P-T389-S6K1 (Rabbit mAb #9234 from Cell signaling). Moreover, the antibody we used in our work, preferentially detects T389-S6K1 in all cell lines and conditions analyzed. Thus, our detection of P-p70-S6K is correct and cannot be attributed to a faulty antibody or misinterpretation of the bands as Siebel et al. conclude.
