1. If your target DNA was in a contaminated sample, make sure you add enough S1 nuclease, you shall adjust its quantity based on your own pre-tests.
2. A good dye can give you a more sensitive result. If you can choose, choose a good dye.
3. It can be used to test expression level by detecting the cDNA, whatever your sample is get from animals, plants, or bacteria, just boil them and add enough enzyme.
