Unfolding was followed by the fluorescence change of SYPRO orange at 570 nm after excitation at 490 nm and measured with 2 μM protein in 50 mM Hepes 7.5, 250 mM NaCl, 100 mM KOAc. The unfolding curves were identical between 0.5 and 5 μM protein.
