An IRF5-dependent pathway is activated locally in the dLN.

WT, Irf3-/-Irf7-/-, or Irf3-/-Irf5-/-Irf7-/- mice were inoculated with PBS (mock) or 103 PFU of CHIKV AF15561 in the left footpad. At 24 hpi, the inoculated foot and the dLN were collected. qRT-PCR was used to analyze mRNA expression of (A) Ccl2, (B) Cxcl10, (C) Ifng, (D) Il6, (E) Ifit1, and (F) Rsad2. Error bars represent mean ± SEM. Data (8–9 mice per group) are combined from 2 independent experiments. Statistical significance was determined by one-way ANOVA with Tukey’s multiple comparisons test. **P < 0.01, ***P < 0.001.
