IL-4/IL-13 polarization of macrophages enhances Ebola virus glycoprotein-dependent infection

A) Analysis of expression of known surface receptors for EBOV in C57BL/6 Ifnar-/- pmacs that were unstimulated or stimulated with 20ng/ml of IL-4/IL-13 for 24 hours. Shown is expression relative to unstimulated controls. B) Expression of SIGNR genes in unstimulated or IL-4/IL-13 stimulated pmacs. Expression was quantified by qRT-PCR and expressed relative to HPRT levels. C) Cell surface expression of EBOV surface receptors in unstimulated or IL-4/IL-13 stimulated cells. Surface expression was evaluated by immunostaining followed by flow cytometry. D) HEK 293T cells were transfected with plasmids expressing the indicated proteins and infected with rVSV/EBOV GP (MOI = 1). Infection was quantified by GFP detection at 24 hours post infection by flow cytometry. Data are shown as mean ± S.D. Each experiment was performed 3 times. Statistical analyses were performed using Student’s t-test, *p<0.05.
