Preferential Elimination of Older Erythrocytes in Circulation and Depressed Bone Marrow Erythropoietic Activity Contribute to Cadmium Induced Anemia in Mice

CD47 expression in different age cohorts of erythrocytes.

Mice were given cadmium chloride dissolved in drinking water (50 ppm and 1000 ppm). Mouse erythrocytes were labeled with biotin in vivo by the two step biotinylation procedure. The experimental protocol is given in panel A. After 13 days of the 2nd biotinylation step, erythrocytes were collected and stained with Streptavidin-APC and anti-mouse CD47-FITC antibody. Representative histograms showing CD47 expression in young and aged erythrocytes is given in panel B. Expression of CD47 on erythrocyte cell membrane among the young (<13 days old) and aged (>35 days old) erythrocytes are shown in panel C. Each bar in the graph represents mean ± SEM of observations on 5 mice. *p<0.005 and **p<0.001 for comparison of the groups (Student t-test).
