Differential Glioma-Associated Tumor Antigen Expression Profiles of Human Glioma Cells Grown in Hypoxia

Hypoxic U251 cells are killed better by Whsc2 specific human CTLs.

HLA-A0201 restricted CTLs were generated in vitro against the Whsc2 103–111 peptide. The U251 target cells were either grown under normoxic or hypoxic conditions for 1 week. The target cells were radio-labeled with Cr51. The lymphocytes were incubated with the target cells for 6 hrs at 37C in quadruplicate cultures. The data is presented as % Specific Release ± Standard Deviation (SD). Asterisks indicate P<0.05 as determined by a Student's t test to show the statistical differences between the cytotoxic data at either the respective 12∶1 or 6∶1 effector:target ratios.
