Ctp1 and the MRN-Complex Are Required for Endonucleolytic Rec12 Removal with Release of a Single Class of Oligonucleotides in Fission Yeast

Rec12-oligonucleotide removal in hop1Δrad50S and mek1Δrad50S depends on Ctp1 and Mre11 nuclease activity.

Rec12myc immunoprecipitates from pat1-114 meiotic time-courses at 4h after temperature shift were analyzed by autoradiography after TdT labeling. Meiotic pat1-114 extracts from the strains hop1Δ ctp1Δ (MR477), mek1Δ rad32-D65N (MR487), wild type (MR388), hop1Δ ctp1Δ rad50S (MR482), and mek1Δ rad32-D65N rad50S (MR484) were immunoprecipitated for Rec12myc and TdT-labeled (upper panel). Western blot analysis revealed presence of Rec12myc (lower panel). The open arrow indicates Rec12myc-oligonucleotide, the filled arrow Rec12myc, and asterisks correspond to non-specific signals.
