A. The model setup is characterized by two different signal contexts (A and ). Cells can reversibly change between A and depending on the cell numbers and the cell specific affinity a. Whereas activated cells in undergo divisions and exponentially degrade their cell specific affinity a, cells in A are quiescent and preserve/regain their affinity a. Further details of the model are given in [14],[17] and in the Supporting Information Text S1. The blue box indicates the region in which cells are considered as HSCs according to a certain purification procedure.
