Calcineurin (gray, catalytic A-subunit; beige, structural B-subunit) is the target of the most widely used immunosuppressant drug, FK506 (ball-and-stick model). FK506 and the viral inhibitor A238L (purple) share a common calcineurin substrate binding site, the LxVP site, which is located at the intersection of the two calcineurin subunits. The structure of the calcineurin:A238L complex led to an in-depth understanding of substrate binding and dephosphorylation by calcineurin. See Grigoriu, et al. (e1001492) in this issue.
